Name ALG5, dolichyl-phosphate beta-glucosyltransferase
Description This gene encodes a member of the glycosyltransferase 2 family. The encoded protein participates in glucosylation of the oligomannose core in N-linked glycosylation of proteins. The addition of glucose residues to the oligomannose core is necessary to ensure substrate recognition, and therefore, effectual transfer of the oligomannose core to the nascent glycoproteins. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2008]

This lipid‐linked glucose donor is essential for the proper assembly of the oligosaccharide precursors required for N‐linked glycosylation in the endoplasmic reticulum. Studies in budding yeast and in the protist Trichomonas vaginalis have demonstrated that ALG5 activity is necessary for efficient protein glycosylation, as its disruption leads to an accumulation of underglucosylated lipid‐linked oligosaccharides and consequent defects in glycoprotein maturation [(1,2,3,4,5,6,7,8)]. In yeast, for example, alg5 mutants show reduced incorporation of glucose into the growing precursor, which in turn diminishes the efficiency with which the glycan is transferred to nascent proteins [(2,3,4,5,6,7,8)]. Likewise, recombinant expression of Trichomonas ALG5 proteins has been shown to restore normal glycosylation in a yeast alg5 mutant, confirming the conserved function of this enzyme in dolichol phosphate–glucose synthesis and its role in the proper glucosylation of both N‐ and, in some cases, O‐linked glycans [(1)]. In addition, human genetic studies have functionally complemented yeast alg5 deficiencies with ALG5 cDNA, underscoring the enzyme’s critical role in glycoprotein biosynthesis and its potential involvement in congenital disorders of glycosylation when mutant or deficient [(9)].
