In the work described, ARLNC1—although referred to as LOC100996425 in the study—is up-regulated in prostate cancer tissues where it directly targets the transcription factor HNF4A. This interaction leads to the suppression of HNF4A expression, which in turn disrupts the balance of the AMPK/mTOR pathway, a critical regulator of cell metabolism, proliferation, and survival. Functionally, overexpression of ARLNC1 promotes cancer cell proliferation and migration, while its knockdown, or restoration of HNF4A levels, significantly reduces these oncogenic properties and induces apoptosis. Moreover, the modulation of the AMPK/mTOR signaling through pharmacologic activators or inhibitors confirms that ARLNC1’s effects are at least partly mediated by this pathway.
