{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n BEX5 is a member of the brain‐expressed X‐linked (BEX) gene family that, although sharing considerable sequence homology and structural features (including long intrinsically disordered regions), appears to have diverged from its paralogs to fulfill a specialized role. Early expression studies showed that human BEX5, identified along with other BEX family members, is localized on the X chromosome and is expressed in the brain, with its protein undergoing proteasome‐mediated degradation (5). Its membership in this family suggests that BEX5 may participate in the regulation of receptor‐mediated signaling events that control processes such as apoptosis and cell cycle regulation, as other family members modulate nerve growth factor (NGF) signaling via interaction with p75NTR and downstream nuclear factor‐κB activation (1, 2). Although functional studies on BEX5 are more limited compared with its counterparts, evolutionary analyses indicate that BEX5 has experienced relatively little gene conversion with the other BEX genes, underscoring a potential acquisition of distinct regulatory roles during mammalian evolution (4). In the cancer context, while the roles of several BEX proteins in modulating cell survival and proliferation have been explored—with some members acting as tumor suppressors and others as oncogenes—the precise functional contribution of BEX5 remained undefined until recent work in uveal melanoma identified high BEX5 expression as a marker associated with better patient prognosis (6).
