This CENP‑S/CENP‑X (alternatively known as the MHF1/MHF2 complex when acting in DNA repair) module plays dual roles. First, in mitosis it is critical for the stable assembly and proper architecture of the outer kinetochore—helping to maintain appropriate kinetochore size and intrakinetochore spacing that are essential for accurate chromosome alignment and segregation. Second, when operating in its alternate guise it partners with the DNA translocase FANCM to facilitate the processing of branched DNA structures and to promote homologous recombination repair at stalled replication forks and interstrand crosslinks.
