ENTREP3 Gene

Name endosomal transmembrane epsin interactor 3
Description This gene is located near the gene for the lysosomal enzyme glucosylceramidase; a deficiency in this enzyme is associated with Gaucher disease. The encoded protein has been identified as a potential binding partner of a WW domain-containing protein which is involved in apoptosis and tumor suppression. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2010]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies in neurological and reproductive contexts have highlighted the critical influence of epigenetic modifications on cellular function. For example, in Parkinson disease models exhibiting levodopa‐induced dyskinesia, pronounced deacetylation of histone H4 and altered acetylation/phosphorylation patterns of histone H3 were observed, suggesting that shifts in chromatin structure can underlie long‐term changes in brain function. In parallel, studies in mouse oocytes have demonstrated that aging and cryopreservation are associated with alterations in acH4K12 levels, implicating such histone modifications in the regulation of developmental potential. Although these investigations do not directly examine ENTREP3, they underscore an overarching biological theme whereby epigenetic mechanisms modulate gene expression and cell behavior—a function that may be shared, at least conceptually, with the regulatory roles attributed to proteins like ENTREP3."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n\nIn a distinct line of research, several studies have identified the oncogenic potential of a protein encoded by the gene Family with Sequence Similarity 189, member B (FAM189B), also known as COTE1—a molecule that in some contexts may be related to or conflated with ENTREP3 function. In hepatocellular carcinoma, COTE1 is markedly upregulated and its overexpression enhances cellular invasion, while its silencing suppresses invasive phenotypes and tumorigenicity. Similar oncogenic roles have been reported in gastric cancer, non‐small cell lung cancer, and intrahepatic cholangiocarcinoma, where elevated COTE1 expression correlates with larger tumor size, advanced clinical stage, and poorer patient survival. Mechanistic studies reveal that COTE1 promotes proliferation and invasion via modulation of key signaling axes, including the suppression of tumor suppressor–mediated pathways involving WWOX (with subsequent effects on the p53 axis), disruption of cyclin D1 regulation, and inhibition of autophagy through downregulation of Beclin1 transcription. Collectively, these observations indicate that ENTREP3/FAM189B/COTE1 functions as a positive regulator of tumor progression by influencing cell cycle progression, apoptotic balance, and autophagic responses, thereby emerging as a promising target for therapeutic intervention."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Anthony P Nicholas, Farah D Lubin, Penelope J Hallett, et al.
