| HGNC Family | WAP four-disulfide core domain containing (WFDC) |
| Name | epididymal peptidase inhibitor |
| Description | This gene encodes an epididymal protease inhibitor, which contains both kunitz-type and WAP-type four-disulfide core (WFDC) protease inhibitor consensus sequences. Most WFDC genes are localized to chromosome 20q12-q13 in two clusters: centromeric and telomeric. This gene is a member of the WFDC gene family and belongs to the telomeric cluster. The protein can inhibit human sperm motility and exhibits antimicrobial activity against E. coli, and polymorphisms in this gene are associated with male infertility. Read-through transcription also exists between this gene and the downstream WFDC6 (WAP four-disulfide core domain 6) gene. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2014] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n Epididymal protease inhibitor (EPPIN) is a multifunctional protein expressed primarily in the testis and epididymis that plays a critical role in male fertility. On the sperm surface, EPPIN binds to seminal plasma proteins—most notably semenogelin‐1—to form a complex that modulates sperm motility by transiently inhibiting progressive movement, an effect that can be reversed by proteolytic digestion during semen liquefaction (1,2,3,17). In addition to its role in regulating motility, EPPIN exhibits antibacterial properties; it disrupts both the outer and inner membranes of bacteria in a manner dependent on intact disulfide bonds and its dual inhibitory domains, suggesting that antimicrobial defense in the male reproductive tract is an additional physiological function (1,4). Moreover, EPPIN may influence intracellular signaling in spermatozoa; antibody‐mediated blockade of EPPIN has been shown to reduce intracellular calcium influx and inhibit the acrosome reaction, thereby demonstrating its regulatory role in fertilization (5). Genetic and immunization studies further support the significance of EPPIN as a key target for non‐hormonal contraception, with specific gene variants correlating with altered semen quality and vaccination against EPPIN resulting in reduced fertility in animal models (6,8,12,13,14,15,16). Collectively, these findings underscore the dual protective and modulatory functions of EPPIN in sperm maturation and function, as well as its promise as a candidate antigen for the development of male contraceptive strategies."}]}, {"type": "t", "text": "\n "}, {"type": "p", "children": [{"type": "b", "children": [{"type": "t", "text": "Citations:"}]}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "17"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Suresh Yenugu, Richard T Richardson, Perumal Sivashanmugam, et al.
