Name fukutin
Description The protein encoded by this gene is a putative transmembrane protein that is localized to the cis-Golgi compartment, where it may be involved in the glycosylation of alpha-dystroglycan in skeletal muscle. The encoded protein is thought to be a glycosyltransferase and could play a role in brain development. Defects in this gene are a cause of Fukuyama-type congenital muscular dystrophy (FCMD), Walker-Warburg syndrome (WWS), limb-girdle muscular dystrophy type 2M (LGMD2M), and dilated cardiomyopathy type 1X (CMD1X). Studies in muscle–eye–brain and Fukuyama‐type congenital muscular dystrophies have shown that fukutin shares homology with fringe‐like glycosyltransferases and is required for the proper elaboration of O‑mannosyl glycans on α‑dystroglycan. In normal cells, fukutin localizes predominantly to the Golgi apparatus where it interacts with other glycosylation enzymes such as POMGnT1, suggesting that it forms part of a functional complex that modulates enzymatic activity in assembling the critical glycan structures. These modifications are crucial because they enable α‑dystroglycan to bind extracellular ligands (for instance, laminin), ensuring basement membrane integrity, proper neuronal migration, and muscle fiber stability. Mutations in FKTN result in hypoglycosylation of α‑dystroglycan with consequent loss of ligand binding, ultimately leading to a range of dystroglycanopathies—from the severe brain and muscle involvement seen in Walker–Warburg syndrome to the milder phenotypes in congenital muscular dystrophies.
