GPRASP3 Gene

Name G protein-coupled receptor associated sorting protein family member 3
Description This gene is a member of a gene family which encodes proteins with a basic helix-loop-helix domain. Other members of this gene family encode proteins which function as transcription factors, either enhancing or inhibiting transcription depending on the activity of other DNA binding proteins. The coding region of this gene is located entirely within the terminal exon. The encoded protein may be involved in the survival of neurons (PMID: 15034937). Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Sep 2011]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nThe protein known as p60TRP, a member of the newly identified basic helix-loop-helix transcription regulator family, plays an important role in modulating neuronal survival and cellular aging. It interacts with key cellular regulators such as Ran-binding-protein-5 and protein-phosphatase-2A, and appears to influence signaling through novel neurotrophin pathways including those mediated by NNT1 and p48ZnF. Its down-regulation in Alzheimer’s disease brains suggests that p60TRP—as a GPCR-associated sorting protein (GPRASP3)—may be pivotal for maintaining the balance between cell survival and programmed cell death in neural tissues."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its neuroprotective functions, p60TRP (GPRASP3) has been implicated in cancer biology. Analyses of human cancer genomes have revealed that distinct regions of the protein are susceptible to specific mutations, with notable hotspots identified at residues E15 and E171. These alterations might affect its role in the broader GPCR signaling cascades—specifically those involving Ras proteins—and underscore its potential relevance as an alternative regulator of G protein signaling in oncogenesis and as a target for new cancer therapeutics."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, the insights from both sets of studies reveal that GPRASP3/p60TRP serves as a critical modulator at the intersection of cell survival, apoptosis, and oncogenic signaling. Its multifaceted interactions and susceptibility to mutation in cancer highlight its dual significance in regulating neurodegenerative processes as well as in tumor biology. This integration of functions underscores the therapeutic potential of targeting GPRASP3 in diverse disease states where cellular signaling and survival are dysregulated."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "K Heese, T Yamada, H Akatsu, et al.
