| Name | H1.8 linker histone |
| Description | Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. The protein encoded is a replication-independent histone that is a member of the histone H1 family. This gene contains introns, unlike most histone genes. The related mouse gene is expressed only in oocytes. [provided by RefSeq, Oct 2015] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n Linker histones of the H1 family—including the oocyte‐specific variant often designated H1FOO (sometimes referred to as H1–8)—play a pivotal role in dynamically organizing chromatin during gametogenesis and early embryogenesis. In mammalian oocytes and early embryos, H1FOO is uniquely expressed and is required for maintaining a relaxed, decondensed chromatin state that facilitates proper transcriptional reprogramming, oocyte maturation, and timely zygotic genome activation. Indeed, functional studies in mice and humans have shown that reduction or misregulation of H1FOO leads to meiotic arrest, impaired polar body formation, aberrant nucleosomal spacing, and ultimately failed embryonic progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "9"}]}, {"type": "t", "text": " Moreover, ectopic expression studies indicate that H1FOO not only sustains a less compact chromatin configuration but can also improve the quality of induced pluripotent stem cells by reducing heterogeneity in differentiation outcomes."}]}, {"type": "t", "text": "\n \n "}, {"type": "p", "children": [{"type": "t", "text": "\n In contrast to the specialized functions of the oocyte‐specific H1 variant, somatic H1 subtypes display diverse genomic distributions and post‐translational modifications that impart context‐dependent roles in modulating chromatin compaction, transcriptional activity, and alternative splicing. For example, certain somatic variants are enriched or depleted at gene promoters and specific chromosomal domains, with some variants even acting as selective repressors of oncogenic noncoding RNAs in cancer cells."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "14"}]}, {"type": "t", "text": " In addition, characterization of H1 profiles in tumors has revealed that lower levels of certain H1 variants correlate with the maintenance of self‐renewing cancer cell states (for instance, via derepression of megabase‐sized gene domains), further underscoring the importance of H1 subtype specialization in cellular identity and function."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "15"}]}, {"type": "t", "text": "\n "}]}, {"type": "t", "text": "\n \n "}, {"type": "p", "children": [{"type": "t", "text": "\n Collectively, these findings underscore that the oocyte‐specific linker histone H1FOO (H1–8) is an essential epigenetic regulator whose precise expression and chromatin binding are critical for proper nuclear remodeling, gene expression control, and developmental competence during the transition from gamete to embryo. At the same time, the functional diversity among somatic H1 variants highlights a broader strategy by which cells fine‐tune higher‐order chromatin organization to regulate genome function in a cell type– and context–dependent manner.\n "}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Mamoru Tanaka, Maki Kihara, Jon D Hennebold, et al.
