H2AC25 Gene

Name H2A clustered histone 25
Description Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H2A family. Transcripts from this gene contain a palindromic termination element. {"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nHIST3H2A, also known as H2AC25, has emerged as an important oncogenic regulator in cancer biology. In non‐small cell lung cancer (NSCLC), reduced expression of miR‑516a‑5p leads to the upregulation of HIST3H2A, which in turn promotes cell viability and anchorage‐independent growth. This deregulation is associated with adverse clinicopathological features, such as larger tumor size and advanced pathological stages, ultimately impacting tumor recurrence and patient survival."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn prostate cancer (PCa), HIST3H2A is significantly overexpressed, where it not only modulates critical cellular processes—including proliferation, migration, invasion, and the epithelial-mesenchymal transition (EMT)—but also specifically regulates programmed necroptosis. Notably, its function in programmed necroptosis is distinct from the apoptosis pathway, underlining its unique role in cancer cell survival and its potential as a therapeutic target."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these studies suggest that HIST3H2A (H2AC25) acts as a multifunctional contributor to tumor progression by interfacing with various molecular pathways. In NSCLC, its elevated expression, driven by the loss of miR‑516a‑5p suppression, enhances tumor cell proliferation, while in PCa, its impact on necroptosis and EMT further promotes an aggressive cancer phenotype. These dual findings underscore the potential of HIST3H2A as both a prognostic marker and a promising target in cancer therapy."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xiang-Yun Ye, Ling Xu, Shun Lu, et al.
