Description Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H2B family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the small histone gene cluster on chromosome 6p22-p21.3. Under normal conditions, H2B family proteins—including H2BC17—are incorporated during S‐phase to package newly replicated DNA and help maintain proper chromatin structure. In addition, post‐translational modifications of H2B proteins regulate nucleosome stability and facilitate dynamic processes such as transcription, DNA repair, and regulated histone exchange. Disruption of these roles by point mutations (for example, alterations analogous to the cancer‐associated H2B E76K mutation) can distort the H2B–H4 interface, induce nucleosome instability, and promote oncogenic transformation by enhancing cell proliferation. Furthermore, evidence indicates that certain replication‐dependent histone genes, including H2BC17, may be expressed as polyadenylated mRNAs in terminally differentiated, nondividing cells where they likely serve as “replacement” histones to sustain chromatin integrity in long‐lived tissues.
