Description Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene encodes a replication-independent histone that is a member of the H2B histone family. [provided by RefSeq, Nov 2015]

Quantitative mass‐spectrometry studies have revealed that H2B isoforms, including H2BW2, are expressed at varying levels across different cell types and cancer lines. Although these isoforms share similar post‐translational modification profiles—for example, comparable acetylation levels—their relative abundance differs in a cell‐type–specific manner. In this context, H2BW2 is thought to contribute to the fine tuning of chromatin structure and epigenetic status, potentially affecting tissue‐specific gene regulation and cellular identity. Furthermore, while the synthesis of canonical histones is tightly coupled to the cell cycle, certain H2B isoforms (including H2BW2) appear to be regulated independently of S‐phase histone expression, suggesting that they might play specialized roles outside of DNA replication.
