| Name | H3 clustered histone 14 |
| Description | Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H3 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in a histone cluster on chromosome 1. This gene is one of four histone genes in the cluster that are duplicated; this record represents the telomeric copy. [provided by RefSeq, Aug 2015] |

Acetylation, methylation, phosphorylation, proteolytic processing, and ubiquitylation of H3 modulate its structure and interactions, thereby governing DNA replication, repair, transcription, and higher order chromatin assembly. For example, acetylation of H3 (e.g. at lysine 56 and 23) promotes timely nucleosome assembly after DNA synthesis and repair by recruiting and coordinating the action of specific histone chaperones such as ASF1, CAF‐1, HIRA, NASP, and DAXX, which in turn ensure correct recycling of parental histones and incorporation of new ones."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": " Variant-specific deposition, as seen with H3.3 and the centromere-specific CENP-A, underlies specialized roles in transcriptional activation, cell reprogramming, and maintenance of centromere identity and kinetochore function."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "13"}]}, {"type": "t", "text": " Moreover, aberrant H3 modification patterns and misregulated histone chaperone activity have been linked to oncogenic processes, where altered H3 acetylation and methylation states drive proliferation, invasion, and metastasis by reprogramming gene expression profiles."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "17"}]}, {"type": "t", "text": " Collectively, these studies demonstrate that through its regulated deposition and modification, histone H3 functions not only as a structural element for packaging the genome but also as an active signal integrator that connects DNA‐templated processes with cellular fate decisions in development and disease.\n "}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Chandrima Das, M Scott Lucia, Kirk C Hansen, et al.
