H4C16 Gene

Name H4 histone 16
Description Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. [provided by RefSeq, Aug 2015]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nHistone H4 acetylation at lysine 16 (H4K16Ac) is emerging as a key regulator of chromatin structure and function. Semisynthetic studies have shown that incorporation of H4K16Ac into nucleosomal arrays disrupts internucleosomal contacts and prevents the formation of compact 30‐nanometer fibers, thereby favoring a more open chromatin conformation that facilitates access of regulatory proteins involved in transcription. In addition, specific chromatin regulatory complexes—such as those containing O‑GlcNAc transferase that target H4K16 along with other H4 lysine residues—further modulate transcription by enhancing histone acetyltransferase activity and maintaining dynamic control over H4K16 acetylation levels."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nH4K16Ac is also implicated in the cellular response to DNA damage and in cancer biology. In tumor samples, diminished H4K16Ac levels have been correlated with increased tumor recurrence and poorer prognosis, suggesting that appropriate H4K16 acetylation plays a tumor‐suppressive role by supporting a chromatin environment less prone to malignant progression. In parallel, studies in DNA double‐strand break repair have revealed that H4K16 acetylation is upregulated upon DNA damage, where its enhanced levels promote chromatin relaxation required for the recruitment of repair factors and efficient repair process activation."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Michael Shogren-Knaak, Haruhiko Ishii, Jian-Min Sun, et al.
