{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\n LAMP5‐AS1 is a long noncoding RNA that appears to play a pivotal role in the pathogenesis of mixed‐lineage leukemia (MLL) by modulating epigenetic regulation. In MLL‐rearranged leukemia, LAMP5‐AS1 is markedly overexpressed and its high expression correlates with reduced leukemia‐free survival. Mechanistic studies have demonstrated that LAMP5‐AS1 directly binds to the lysine‐rich region within the catalytic domain of the histone methyltransferase DOT1L, thereby enhancing DOT1L’s activity. This interaction leads to elevated levels of global H3K79 di‐ and tri‐methylation, which in turn upregulates the expression of self‐renewal genes in the HOXA cluster that are critical for maintaining the stemness of leukemia cells. Functionally, suppression of LAMP5‐AS1 significantly reduces colony formation and promotes differentiation of primary MLL leukemia CD34+ cells, highlighting its potential as a therapeutic target in this aggressive leukemia subtype."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n "}]}, {"type": "t", "text": "\n "}, {"type": "p", "children": [{"type": "t", "text": "\n In a broader context of long noncoding RNA studies in cancer, additional work has explored ceRNA regulatory networks in pediatric malignancies such as Wilms tumor, wherein several differentially expressed lncRNAs have been associated with tumor stage and overall survival. However, among these studies, LAMP5‐AS1 has not been specifically characterized outside the setting of MLL‐rearranged leukemia."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n "}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Wen-Tao Wang, Tian-Qi Chen, Zhan-Cheng Zeng, et al.
