In colon cancer tissues, LINC00538 is significantly overexpressed relative to matched normal tissues and its high expression correlates with poorer overall survival. Functional assays in colon cancer cell lines (e.g., HCT116 and RKO) have shown that silencing LINC00538 attenuates cell proliferation, shortens the S phase of the cell cycle, and induces apoptosis. Mechanistic studies further revealed that LINC00538 exerts its tumor‐promoting effects by binding to the epigenetic regulator EZH2, which in turn leads to the suppression of NKD2 expression—a change that appears to facilitate unchecked cell proliferation and survival.
