| Name | microRNA 105-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‐105 has emerged as a multifaceted regulator in cancer biology. In the tumor microenvironment, cancer cells—most notably in breast cancer—secrete miR‐105 via extracellular vesicles to reprogram stromal fibroblasts. This reprogramming alters cellular metabolism, enhancing nutrient utilization and waste detoxification to sustain tumor growth. In parallel, miR‐105 produced as part of a cancer‐germline transcript has been shown to disrupt vascular endothelial barriers, thereby facilitating metastasis, while similar extracellular vesicle–mediated mechanisms re‐prioritize mRNA translation pathways under nutrient fluctuations."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its effects on the tumor niche, miR‐105 can function in a cell‐intrinsic manner as a tumor suppressor in several cancers. In prostate cancer, for instance, reduced levels of miR‐105 are associated with enhanced proliferation and invasiveness, with its restoration leading to significant inhibition of oncogenic properties. Similar tumor‐suppressive roles are observed in gastric and esophageal carcinomas, and in specific breast and ovarian cancers, where diminished miR‐105 expression correlates with aggressive disease and chemoresistance. Mechanistically, miR‐105 targets critical mediators such as CDK6, SOX9, and ZNRF2, thereby modulating cell cycle progression, differentiation, and drug sensitivity."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its oncogenic and tumor‐suppressive roles, miR‐105 also influences differentiation pathways in both normal and diseased states. It acts as a potent enhancer of megakaryopoiesis during hematopoietic differentiation by targeting transcription factors such as c‐Myb. In stem cell models, miR‐105 promotes osteogenic differentiation through modulation of SOX9, and in osteosarcoma, its 5p‐variant is integrated into a regulatory axis—modulated by circular RNAs—that controls FOXO1 expression, affecting cell cycle arrest and apoptosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "12"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Axelle Loriot, Aurélie Van Tongelen, Jordi Blanco, et al.
