MIR1185-1 Gene

HGNC Family Non-coding RNAs
Name microRNA 1185-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nIn colorectal cancer, MIR1185-1 acts as a novel tumor suppressor microRNA that is epigenetically regulated by SIRT1. SIRT1 inhibition leads to enhanced histone acetylation on the MIR1185-1 promoter, increasing its expression. Elevated MIR1185-1 subsequently targets the 3′UTR of the CD24 mRNA, downregulating CD24 expression and thereby reducing cancer stemness properties and tumor growth. This SIRT1–MIR1185-1–CD24 regulatory axis highlights the potential of MIR1185-1 as a therapeutic target in colorectal cancer."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn the context of metabolic regulation, MIR1185-1 has been identified as a biomarker linked to weight-loss response. In a study comparing high responders to low responders in a dietary intervention, MIR1185-1 levels were significantly elevated in high responders. Functional assays demonstrated that MIR1185-1 binds to the 3′UTR of GSK3B, leading to diminished expression of this proinflammatory gene, suggesting a role for MIR1185-1 in modulating inflammatory processes associated with metabolic improvement upon weight loss."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, MIR1185-1 is implicated in the neurobiology of schizophrenia through its posttranscriptional regulation of glial cell line-derived neurotrophic factor (GDNF). A risk-associated single nucleotide polymorphism (SNP) in the 3′UTR of the GDNF gene creates a binding site for MIR1185-1, and in vitro studies have shown that this microRNA represses reporter activity in a dose-dependent manner when the risk allele is present. This finding suggests that MIR1185-1 may contribute to the downregulation of GDNF expression in patients with schizophrenia, potentially impacting dopaminergic neuron survival and function."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Teh-Wei Wang, Edward Chern, Chao-Wei Hsu, et al.
