| HGNC Family | Non-coding RNAs |
| Name | microRNA 1204 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nRecent studies have revealed that miR-1204, a microRNA embedded within the large non-coding PVT1 locus, plays versatile roles in cancer biology. Initially, it was shown that p53, a key tumor suppressor, directly binds to and activates regulatory elements within the PVT1 locus, thereby upregulating miR-1204 among other non-coding RNAs. Notably, ectopic miR-1204 expression was found to elevate p53 levels and trigger cell death in a partially p53-dependent manner, suggesting the existence of a regulatory feedback loop linking p53 activity and miR-1204 expression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nSubsequent investigations have uncovered multiple oncogenic roles for miR-1204 depending on the tumor context. In nasopharyngeal carcinoma, miR-1204 is notably downregulated in paclitaxel-resistant cells, and its restoration re-sensitizes these cells to chemotherapy while inhibiting tumor growth, indicating its potential utility in overcoming drug resistance. In breast cancer, increased miR-1204 expression correlates with poor prognosis; it drives proliferation, epithelial-mesenchymal transition, and invasion by targeting the vitamin D receptor (VDR). Similarly, in ovarian squamous cell carcinoma, miR-1204 overexpression promotes tumor growth by upregulating GLUT-1 expression and enhancing glucose uptake. In non-small-cell lung cancer, elevated miR-1204 levels contribute to enhanced cell proliferation through downregulation of the tumor suppressor PITX1, and additional studies are investigating its influence on DEK oncogene expression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "2", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, the role of miR-1204 in colorectal cancer remains to be clarified, despite its presence in the PVT1 locus which has been implicated in diverse tumorigenic processes.
