| HGNC Family | Non-coding RNAs |
| Name | microRNA 1207 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‐1207 (including its 5p and 3p forms) is emerging as a critical regulator in cancer biology, where its expression and function vary according to tumor type. In gastric cancer, for example, one study showed that an overexpressed lncRNA (BC032469) acts as a competing endogenous RNA by sponging miR‐1207‐5p, thereby relieving hTERT from miRNA‐mediated suppression, while another report directly demonstrated that miR‐1207‐5p targets the hTERT 3′ UTR to inhibit tumor growth. Similar context‐dependent roles have been noted in other cancers: in breast cancer, a PVT1‐derived miR‐1207‐5p promotes cell proliferation by downregulating STAT6 and affecting cell cycle regulators; in lung cancer, miR‐1207‐5p inhibits epithelial–mesenchymal transition (EMT) and metastasis through direct targeting of CSF1, which consequently dampens STAT3 and AKT signaling; and in prostate cancer, the miR‐1207‐3p isoform has been reported to function as a tumor suppressor by targeting FNDC1, leading to decreased activity of fibronectin and androgen receptor pathways. In ovarian cancer, miR‐1207 overexpression facilitates cancer stem cell–like properties by targeting multiple negative regulators of the Wnt/β‐catenin pathway, while in colorectal and nasopharyngeal carcinomas, deregulation of miR‐1207‐5p—often through sequestration by circular RNAs or long non‐coding RNAs (lncRNAs)—influences cell proliferation, invasion, and stemness."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumor cell proliferation, invasion and metastasis, miR‐1207 also participates in non‐cancerous pathologies. In diabetic nephropathy, for example, miR‐1207‐5p is upregulated by high glucose and transforming growth factor–β1 (TGF‐β1) and contributes to extracellular matrix accumulation by enhancing the expression of TGF‐β1, plasminogen activator inhibitor 1, and fibronectin. In podocytes, a naturally occurring single nucleotide polymorphism in the HBEGF 3′UTR disrupts miR‐1207‐5p binding, leading to altered HBEGF regulation. Moreover, in the context of infectious and inflammatory diseases, miR‐1207‐5p has been implicated in SARS‐CoV‐2 infection by regulating CSF1 expression, a modulator of macrophage-mediated inflammation, and its aberrant expression has been associated with autoimmune conditions such as Sjögren’s syndrome, where downregulation of miR‐1207‐5p correlates with increased TRIM21 levels and enhanced apoptosis. Several studies have demonstrated that lncRNAs (such as PVT1, LINC00319, LUADT1, and PTV1) and circular RNAs (such as circHIPK3) bind miR‐1207‐5p, thereby modulating its ability to target downstream effectors including hTERT, STAT6, KLF12, SEPT9, and components of the Wnt/β‐catenin, Akt/mTOR, and PI3K/AKT pathways. In prostate cancer, for instance, miR‐1207‐3p targets PYCR1, curtailing cell proliferation, while in cervical carcinoma, LUADT1 binds miR‐1207‐5p to upregulate SEPT9 and promote tumor growth. In addition, hnRNPK‐regulated antisense transcripts can sequester miR‐1207‐5p, leading to modulation of heme oxygenase–1 (HO‐1) and impacting cell survival.
