MIR1224 Gene

HGNC Family Non-coding RNAs
Name microRNA 1224
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‑1224‑5p is emerging as a multifunctional regulator in cancer, acting predominantly as a tumor suppressor in several solid malignancies. In high‑grade gliomas, its reduced expression contributes to enhanced proliferation and invasion while its restoration promotes apoptosis via direct inhibition of CREB1 (#1). In keloids, overexpression of miR‑1224‑5p in fibroblasts suppresses proliferation and migration and triggers apoptosis, suggesting a role in limiting abnormal scar formation (#4). In lung cancer, an interplay with lncRNA NEAT1 implies that miR‑1224‑5p may influence tumor progression (#8). Downregulation of miR‑1224‑5p in laryngeal papillomas is linked to increased OGFOD1 levels and consequent cell survival (#9). In glioblastoma, miR‑1224‑5p is sequestered by the lncRNA MIR4435‑2HG, relieving its inhibition of TGFBR2 and thereby promoting oncogenic signaling (#10). Furthermore, in rectal cancer, miR‑1224‑5p directly targets SLC29A3 to reduce cell viability, invasion, and migration (#11), while in oral squamous cell carcinoma, exosomal transfer of lncRNA APCDD1L‑AS1 diminishes miR‑1224‑5p levels to modulate NSD2 expression and confer chemoresistance (#12)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond cancer, miR‑1224‑5p plays divergent roles in metabolic and inflammatory contexts. Its function has been initially explored in acute liver failure, although the precise mechanisms remain to be elucidated (#2). In the liver, miR‑1224‑5p promotes lipogenesis by directly targeting the AMPKα1 subunit, thereby increasing triglyceride accumulation and contributing to steatosis (#5). During Bacillus Calmette‑Guérin infection, miR‑1224 expression is markedly induced in macrophages, suggesting a role in host immune responses (#6). Its contribution in hepatocellular carcinoma tumor endothelial cells is currently under investigation (#7). In polycystic ovary syndrome, upregulation of miR‑1224‑5p dampens TNF‑α‑induced secretion of androgens and attenuates inflammatory signaling via repression of FOXO1 and subsequent NLRP3 inflammasome activation (#13). Moreover, in sepsis‑related acute lung injury, miR‑1224‑5p aggravates inflammation and oxidative stress by inhibiting the PPAR‑γ/AMPKα axis (#14)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMiR‑1224‑5p also modulates vascular functions. In endothelial cells, its upregulation enhances capillary tube formation by boosting VEGF signaling and concurrently repressing NOTCH signaling through direct targeting of the endocytosis adaptor epsin2 (#3). In diabetic retinopathy, the SPAG5‑AS1/miR‑1224‑5p/IRS‑1 regulatory axis has been shown to influence retinal microvascular endothelial cell behavior under hyperglycemic conditions, suggesting therapeutic potential (#15). Furthermore, circRSU1 acts as a molecular sponge for miR‑1224‑5p to control ITGA5 expression, thereby modulating endothelial cell permeability and function in the setting of septic acute lung injury (#16)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jin Qian, Rui Li, Ying-Yi Wang, et al.
