| HGNC Family | Non-coding RNAs |
| Name | microRNA 1244-1 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Restoration of miR‑1244 levels suppresses cell viability, promotes caspase‑3 activity, and upregulates apoptotic mediators (p53 and Bax), while downregulating proliferative factors such as MEF2D and cyclin D1. Together, these findings indicate that miR‑1244 functions as a tumor suppressor by enhancing chemosensitivity and promoting apoptosis in lung cancer cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn osteosarcoma, a circRNA—circSAMD4—has been shown to be highly expressed in tumor tissues relative to adjacent non‐cancerous tissues. The circSAMD4/miR‑1244/MDM2 regulatory loop promotes cell proliferation and enhances stemness features in osteosarcoma cells."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn ovarian cancer, miR‑1244 activity is similarly subverted by non‑coding RNAs. LINC00504, a long non‑coding RNA, acts as an endogenous sponge to downregulate miR‑1244, consequently facilitating increased cell proliferation, reduced apoptosis, and a metabolic shift toward aerobic glycolysis via modulation of key glycolytic enzymes (PKM2, HK2, and PDK1). In parallel, the circular RNA circ_0002711 sequesters miR‑1244 to relieve its suppression on ROCK1, further promoting tumor growth and the Warburg effect.
