| HGNC Family | Non-coding RNAs |
| Name | microRNA 1249 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐1249 plays multifaceted roles in cancer by functioning largely as a tumor suppressor. In colorectal cancer, its marked downregulation—owing in part to its induction by p53—leads to increased expression of VEGFA and HMGA2, thereby promoting tumor growth, metastasis, angiogenesis, and epithelial–mesenchymal transition via Akt/mTOR signaling. Similarly, in colon adenocarcinoma, decreased miR‐1249 expression correlates with poor prognosis, and in hepatocellular carcinoma, aberrant levels of miR‐1249‐3p have been documented, implicating it in hepatocarcinogenesis even though its mechanistic details remain to be fully deciphered."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncogenesis, miR‐1249 contributes to antiviral defense and bone biology. It directly targets the PB2 coding region in influenza A viruses, effectively suppressing viral replication of both H5N1 and H1N1 strains, which underscores its role in the host’s innate antiviral response. In a different biological context, miR‐1249‐5p has been shown to promote osteogenic differentiation of adipose‐derived stem cells, suggesting therapeutic potential in conditions such as osteoporosis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "4"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn lung cancers and normal epithelial cells, regulatory networks involving miR‐1249‐3p have been elucidated through its interactions with specific circular RNAs and transcription factors. In lung adenocarcinoma, circ_0010235 acts as an oncogenic sponge for miR‐1249‐3p, leading to the upregulation of HOXA13 and enhancement of malignant behaviors including proliferation, migration, invasion, and EMT. A parallel mechanism is observed in non‐small cell lung cancer, where circ_0001715 sequesters miR‐1249‐3p to elevate FGF5 expression and promote tumor progression. Moreover, in normal epithelial cells, inhibition of miR‐1249‐3p results in increased HOXA13 and downstream β‐catenin expression, which suggests a role for miR‐1249 in modulating cell adhesion."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "8"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Xiaoxiang Chen, Kaixuan Zeng, Mu Xu, et al.
