| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In non‑Hodgkin’s lymphoma, miR‑1250 (specifically the miR‑1250‑5p form) is embedded in the second intron of the AATK gene on chromosome 17q25. The adjacent CpG island in the promoter region of AATK suggests that both the host gene and its intronic miRNA are co‑regulated via promoter DNA methylation, supporting a tumor suppressor role for miR‑1250‑5p in this malignancy."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn another study focused on colorectal cancer, the authors explored the influence of long non‑coding RNAs on tumorigenesis through a miRNA sponging mechanism. Although this work centered on lncRNA ELFN1‑AS1 without directly addressing miR‑1250, it underscores the broader paradigm whereby lncRNAs can modulate gene expression by sequestering specific microRNAs. This mechanism offers a conceptual framework that could be applicable to understanding miR‑1250 regulation in cancer contexts."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nFurther complexity in miR‑1250 function emerges from studies in diabetic nephropathy, where the miR‑1250‑3p variant plays a critical regulatory role. Here, miR‑1250‑3p normally binds to and downregulates MSH2, a key mediator in cellular proliferation and fibrosis. The long non‑coding RNA LINC01232, overexpressed under diabetic conditions, acts as a molecular decoy by sequestering miR‑1250‑3p. This sequestration relieves the suppression on MSH2, leading to its increased expression and, correspondingly, the alleviation of mesangial cell proliferation and fibrosis in diabetic kidney models."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Min Yue Zhang, Lu Qian Wang, Chor Sang Chim "}, {"type": "b", "children": [{"type": "t", "text": "miR-1250-5p is a novel tumor suppressive intronic miRNA hypermethylated in non-Hodgkin's lymphoma: novel targets with impact on ERK signaling and cell migration."}]}, {"type": "t", "text": " "}, {"type": "i", "children": [{"type": "t", "text": "Cell Commun Signal (2021)"}]}, {"type": "t", "text": " DOI: "}, {"type": "a", "children": [{"type": "t", "text": "10.1186/s12964-021-00707-0"}], "href": "https://doi.org/10.1186/s12964-021-00707-0"}, {"type": "t", "text": " PMID: "}, {"type": "a", "children": [{"type": "t", "text": "34044822"}], "href": "https://pubmed.ncbi.nlm.nih.gov/34044822"}]}, {"type": "r", "ref": 2, "children": [{"type": "t", "text": "L-Q Zhai, X-X Wang, C-X Qu, et al.
