| HGNC Family | Non-coding RNAs |
| Name | microRNA 1253 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nExtensive research across multiple cancer types has revealed that miR‑1253 primarily functions as a tumor suppressor. In pancreatic ductal adenocarcinoma, for example, the upregulated circular RNA circ_0030235 was shown to exert its oncogenic effects partly by sponging miR‑1253, thereby diminishing its regulatory influence on downstream targets. Similarly, in non‑small‑cell lung carcinoma, miR‑1253 is found significantly down‑regulated, and its restoration suppresses cell proliferation, migration, and invasion through the direct targeting of the WNT5A (long) isoform. In medulloblastoma, epigenetic silencing via hypermethylation reduces miR‑1253 expression, facilitating tumor cell proliferation by failing to repress oncogenic targets such as CDK6 and CD276."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn additional tumor contexts, similar tumor‑suppressive roles for miR‑1253 have been described. In osteosarcoma, the circular RNA circNASP sequesters miR‑1253, which normally targets FOXF1; this sequestration leads to enhanced cell proliferation and invasion. In prostate and colon cancers, miR‑1253 downregulation correlates with advanced disease and poor prognosis, acting through the regulation of EZH2 expression to impair oncogenic processes. Although its role in osteosarcoma is not yet fully defined, these findings collectively indicate that the attenuation of miR‑1253 activity by mechanisms such as ceRNA sponging or epigenetic silencing can facilitate tumor progression. Its emerging involvement—even in studies addressing chemoresistance in non‑small‑cell lung cancer—positions miR‑1253 as a promising prognostic biomarker and potential therapeutic target."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Meiyue Liu, Yue Zhang, Jie Zhang, et al.
