MIR1258 Gene

HGNC Family Non-coding RNAs
Name microRNA 1258
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐1258 is a small noncoding RNA that has emerged as a crucial tumor‐suppressive regulator across a broad spectrum of cancers. Aberrant downregulation of miR‐1258 has been documented in colorectal cancer, non‐small cell lung cancer, hepatocellular carcinoma, osteosarcoma, and other malignancies, where its diminished expression is associated with enhanced tumor growth, invasion, metastasis, and poorer clinical outcomes. These observations indicate that miR‐1258 may serve as an important biomarker for disease progression and prognosis in diverse oncologic settings."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistically, miR‐1258 acts by directly targeting a range of key molecules that drive oncogenic processes. In non‐small cell lung cancer and breast cancer, for example, it negatively regulates heparanase (HPSE) expression, while in Kaposi’s sarcoma–associated herpesvirus infection it targets the viral major lytic switch protein RTA to promote viral latency. Additionally, miR‐1258 suppresses the expression of cyclin‐dependent kinase regulatory subunit 1B (CKS1B) in colorectal and liver cancers, downregulates AKT3 in osteosarcoma, and targets transcription factors such as E2F1 in cervical and breast cancers and SP1 in oral squamous cell carcinoma. Epigenetic alterations—such as hypermethylation of the miR‐1258 promoter—have also been implicated in its silencing in tumors, thereby mitigating its ability to restrain these oncogenic signaling cascades and contributing to tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in tumor suppression, miR‐1258 is also involved in other biological processes. In the context of viral oncogenesis, miR‐1258 contributes to maintaining latency by suppressing lytic gene expression, thereby influencing the aggressive behavior of AIDS‐related malignancies. Furthermore, in septic acute lung injury, miR‐1258 modulates the TGF‑β1/SMAD3 pathway through its targeting of Prep1 to mitigate oxidative stress and inflammation. Emerging evidence also hints at a complex interplay between miR‐1258 and other noncoding RNAs—such as the recently characterized circ_0046600 in hepatocellular carcinoma—that may further modulate cell migratory behaviors and tumor dynamics, underscoring the multifaceted roles of miR‐1258 in both cancer and inflammatory pathologies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Hongcheng Liu, Xiaofeng Chen, Wen Gao, et al.
