MIR1301 Gene

HGNC Family Non-coding RNAs
Name microRNA 1301
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‑1301‑3p plays context‐dependent roles across a spectrum of human diseases, particularly in cancer. In some malignancies it exhibits oncogenic behavior—for example, in prostate cancer, miR‑1301‑3p promotes cancer stem cell expansion by directly repressing Wnt pathway inhibitors such as GSK3β and SFRP1, and further drives cell cycle progression by downregulating tumor suppressors like PPP2R2C. In non‐small cell lung cancer, elevated levels of miR‑1301‑3p enhance proliferation, migration, invasion, and even radioresistance through inhibition of the homeodomain‐only protein HOPX. Conversely, in hepatocellular carcinoma, osteosarcoma, colorectal cancer, and triple‐negative breast cancer, miR‑1301‑3p acts as a tumor suppressor by targeting oncogenic factors including UBE4B, BCL9, and STAT3 and by potentiating p53 activity, thereby inhibiting cell growth and metastatic behavior."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "8"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its intrinsic activity, miR‑1301‑3p is subject to regulation by a complex network of long non‑coding RNAs that function as competing endogenous RNAs. For instance, in osteosarcoma the lncRNA SNHG16 sequesters miR‑1301‑3p, leading to increased expression of BCL9, whereas in esophageal squamous cell carcinoma the lncRNA MIAT binds miR‑1301‑3p to de‐repress INCENP, thereby promoting proliferation and cell cycle progression. Similarly, in hepatocellular carcinoma, lncRNA LINC01433 suppresses miR‑1301‑3p to enable STAT3 upregulation, and in glioblastoma and diabetic retinopathy, other lncRNAs (including PVT1 and those yet to be fully characterized) modulate miR‑1301‑3p activity to affect targets such as TMBIM6 and KLF7. Moreover, in bone marrow mesenchymal stem cells, lncRNA DANCR sponges miR‑1301‑3p to relieve its inhibition of PROX1, thereby suppressing osteogenic differentiation. Preliminary studies in ovarian cancer further suggest that miR‑1301 may regulate cisplatin sensitivity, although its precise role in chemoresistance remains to be clarified."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "9", "end_ref": "18"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its effects on cell proliferation and survival, miR‑1301‑3p modulates key signaling cascades and therapeutic responses. For example, in squamous cell carcinoma models, overexpression of miR‑1301‑3p results in significant downregulation of PI3K and reduced AKT phosphorylation, implicating inhibition of the PI3K/AKT pathway in its antitumor actions. In chronic myeloid leukemia, downregulation of RanGAP1 by miR‑1301 leads to nuclear entrapment of the oncogenic BCR‑ABL fusion protein and thereby enhances p73‐dependent apoptosis, which augments the efficacy of imatinib treatment. On the other hand, in non‑small cell lung cancer, heightened miR‑1301‑3p activity drives tumor progression and radioresistance via suppression of HOPX."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "19", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Lin Fang, Ning Yang, Jie Ma, et al.
