| HGNC Family | Non-coding RNAs |
| Name | microRNA 133a-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR133A2 is a member of the miR‐133 family that plays a multifaceted role in cancer biology. In numerous studies its downregulation in malignant tissues correlates with more aggressive phenotypes and poorer patient outcomes. In colorectal, lung, gastric, cervical, ovarian, pancreatic, renal, glioma and breast cancers, MIR133A2 (often described along with its family members) acts as a tumor suppressor by repressing the expression of key oncogenes and pro‐metastatic signaling molecules – for example, by interfering with pathways involving GEF‐H1/RhoA, TAGLN2, ARPC5, GSTP1, CTGF, IGF1R, TGFBR1, YES1, and RGS3. In these contexts, restoration of MIR133A2 levels suppresses cell proliferation, invasion, and metastatic spread, whereas its epigenetic silencing (for instance via aberrant promoter methylation) or sequestration by oncogenic long non‐coding RNAs contributes to tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its oncogenic roles, MIR133A2 is pivotal in the regulation of cardiovascular and other non‐malignant processes. In myocardial infarction and in conditions of atrial fibrillation, its expression in heart tissue is closely linked to proper electrical conduction and contractile function. In primary human lung fibroblasts, MIR133A2 is induced by transforming growth factor–β1 and then acts as an anti‐fibrotic factor by repressing profibrogenic genes, thus mediating a negative feedback loop to limit myofibroblast differentiation. In endothelial cells, MIR133A2 (especially the 3p strand) directly modulates the levels of key proteins such as aquaporin‑1, which contributes to endothelial migration, tube formation, and overall angiogenic capacity. Moreover, its circulating levels have been associated with biomarkers of vascular injury and early end‐organ damage in conditions like hypertension, further supporting a role in cardiovascular homeostasis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "23"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yasumitsu Moriya, Nijiro Nohata, Takashi Kinoshita, et al.
