| HGNC Family | Non-coding RNAs |
| Name | microRNA 1469 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In inflammatory skin conditions such as acne, keratinocytes exposed to biofilm-derived Cutibacterium acnes exhibit an upregulation of miR-146a following toll-like receptor 2 (TLR2) activation. This miRNA directly targets and suppresses the expression of adapter molecules IRAK1 and TRAF6, thereby attenuating downstream NF‑κB and MAPK signaling cascades and ultimately reducing the secretion of proinflammatory cytokines IL‑6, IL‑8, and TNF‑α. These findings highlight a predominantly anti‑inflammatory role in the regulation of acne pathogenesis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn contrast, in the context of pancreatic cancer, the miR‑1469‑5p isoform is significantly upregulated, with elevated levels correlating with lymph node metastasis and advanced TNM stage. Functional analyses indicate that miR‑1469‑5p exerts oncogenic effects by directly binding to and downregulating the metastasis suppressor NDRG1, which in turn leads to the activation of the NF‑κB pathway and reduction of the cell adhesion molecule E‑cadherin. This molecular axis promotes cancer cell proliferation and invasion, suggesting that miR‑1469‑5p could serve as a prognostic biomarker and potential therapeutic target in pancreatic malignancies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nConversely, in primary cutaneous melanoma, miR‑1469 is expressed at lower levels in ulcerated tumors compared to non‑ulcerated tumors. Restoration of miR‑1469 through mimic transfection resulted in a marked decrease in melanoma cell migratory and invasive capacities, an effect at least partly mediated by reduced expression of its target gene MCL1. These data imply that miR‑1469 functions as a tumor suppressor in melanoma by specifically modulating pathways that underlie cellular invasion and migration, thereby influencing disease progression."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Rong Zeng, Haoxiang Xu, Yuzhen Liu, et al.
