MIR147A Gene

HGNC Family Non-coding RNAs
Name microRNA 147a
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA MIR147A has emerged as an important tumor‐suppressive regulator in several cancer types. In breast cancer, its co‐regulation of EGFR‐driven cell‐cycle network proteins highlights its capacity to inhibit cell proliferation, while in chronic myeloid leukemia, its interaction with the lncRNA MEG3 suggests a role in modulating malignant growth. In colon cancer, overexpression of MIR147A reduces stem cell–like traits by suppressing epithelial–mesenchymal transition through regulation of the Wnt/β‐catenin pathway, and similar tumor‐inhibitory properties are observed in non‐small‐cell lung cancer and oral squamous cell carcinoma by targeting oncogenic mediators such as IRX5. In nasopharyngeal carcinoma, the circSETD3/miR-147a axis modulates Akt/mTOR signaling to counteract cisplatin resistance."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its oncogenic roles, MIR147A is dynamically regulated under stress and inflammatory conditions. Under hypoxia, HIF‐1α upregulates MIR147A, which in turn stabilizes HIF‐1α via targeting HIF‐3α, forming a positive feedback loop that contributes to growth arrest. In glioblastoma, MIR147A modulates ferroptotic cell death, while its elevated expression in progressive supranuclear palsy is associated with repression of genes critical for cellular homeostasis. Furthermore, an extensive review has highlighted its role in regulating macrophage function via TLR4 signaling, and exosome‐mediated delivery of MIR147A can alleviate pathological angiogenesis and inflammatory injury in atopic dermatitis. In the vascular context, MIR147A also targets ZEB2 to reduce monocyte adherence to endothelial cells, thereby potentially mitigating atherosclerotic plaque formation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "7", "end_ref": "12"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these findings underscore the multifaceted functions of MIR147A as a molecular rheostat in disease pathogenesis. Its capacity to target diverse effectors—including HIF-3α, VEGFA, ZEB2, and IRX5—enables MIR147A to modulate key aspects of cellular proliferation, differentiation, stress responses, and inflammatory signaling across a variety of pathophysiological contexts. Such convergence of actions makes MIR147A a promising candidate for therapeutic intervention in cancers, neurodegenerative disorders, and inflammatory as well as vascular diseases."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}, {"type": "fg_f", "ref": "7"}, {"type": "fg_f", "ref": "4"}, {"type": "fg_fs", "start_ref": "8", "end_ref": "11"}, {"type": "fg_f", "ref": "5"}, {"type": "fg_f", "ref": "12"}, {"type": "fg_f", "ref": "6"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Stefan Uhlmann, Heiko Mannsperger, Jitao David Zhang, et al.
