| HGNC Family | Non-coding RNAs |
| Name | microRNA 150 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐150 is a versatile, small non‐coding RNA that plays essential roles in normal hematopoiesis and intercellular communication. In developmental models and immune cells, miR‐150 has been shown to directly target transcription factors such as c‐Myb—thereby modulating cell differentiation in zebrafish and human hematopoietic lineages—and to become strongly up‐regulated during T‐cell maturation where it suppresses Notch3, ultimately influencing proliferation and survival. Moreover, miR‐150 can be actively secreted in microvesicles from monocytes, acting in a paracrine manner to promote endothelial cell migration, while its aberrant expression in natural killer/T‐cell malignancies underscores its tumor–suppressive activities in certain lymphoid contexts."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn cancer settings, miR‐150 exhibits context‐dependent functions. In certain tumors, such as gastric cancer, its overexpression promotes tumorigenesis via direct targeting of tumor–suppressor genes (eg, EGR2), whereas in others—including pancreatic, colorectal, hepatocellular, esophageal, ovarian and some forms of leukemia and lung cancer—miR‐150 acts as a tumor suppressor by downregulating key oncogenes and mediators of epithelial–mesenchymal transition (eg, MUC4, ZEB1, VEGFA, MMP14, FOXO4). In these cancers its expression is often perturbed by competing endogenous RNAs such as lncRNAs (ZFAS1, CASC11) or circular RNAs (eg, cir‐ZNF609), which sequester miR‐150 and thereby abrogate its regulatory functions. These studies collectively indicate that restoring miR‐150 levels or interfering with its upstream inhibitory networks may represent novel therapeutic strategies."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "21"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles within cells, miR‐150 is emerging as an important circulating biomarker in several non–neoplastic conditions. In fibrotic disorders such as lupus nephritis, miR‐150 contributes to pathological extracellular matrix deposition by targeting antifibrotic mediators (eg, SOCS1).
