| HGNC Family | Non-coding RNAs |
| Name | microRNA 15a |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Multiple independent studies have demonstrated that loss of MIR15A expression contributes to tumorigenesis by impairing pro‐apoptotic responses and deregulating cell cycle progression. In CLL, for example, MIR15A (together with MIR16–1) normally functions to negatively regulate the anti‐apoptotic protein BCL2, thereby promoting apoptosis of malignant B cells. This tumor‐suppressive role has also been observed in other cancers, where reduced MIR15A levels are associated with increased cellular proliferation and survival."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "10"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMechanistic studies reveal that MIR15A exerts its tumor‐suppressive effect by directly targeting critical oncogenic transcripts. Beyond repressing BCL2, MIR15A modulates the expression of several genes involved in cell‐cycle control—including cyclin D1 and MCL1—as well as pathways controlling apoptosis, proliferation, and even angiogenesis. Additional research has identified that MIR15A is embedded in complex regulatory networks involving transcription factors such as E2F1 and c-Myc, which govern its transcriptional regulation. These multifaceted interactions underscore the role of MIR15A not only in promoting programmed cell death but also in ensuring proper cell cycle exit."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "11", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nClinically, down-regulation of MIR15A has been closely linked to disease progression, chemoresistance, and metastasis. Its loss not only contributes to an impaired apoptotic response and deregulated cell proliferation but also affects the tumor microenvironment—impacting processes such as angiogenesis and drug sensitivity. As a consequence, experimental restoration of MIR15A levels has been shown to resensitize cancer cells to apoptosis and inhibit tumor growth and invasion in a variety of models, highlighting its potential as a therapeutic target.
