MIR15B Gene

HGNC Family Non-coding RNAs
Name microRNA 15b
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐15b (miR‐15b) is emerging as a multifunctional regulator in diverse cancers, where its role appears highly context dependent. In several malignancies—including tongue squamous cell carcinoma, glioma, neuroblastoma, hepatocellular carcinoma, liver cancer, osteosarcoma, colorectal cancer, and B‐cell chronic lymphocytic leukemia—miR‐15b functions as a tumor suppressor by promoting cell cycle arrest, apoptosis, and inhibition of epithelial–mesenchymal transition through targeting oncogenic drivers such as BMI1, cyclin E1, MYCN, Bcl‐w, Rab1A, DCLK1, and others. In contrast, in prostate and certain gastric cancers, aberrantly high levels of miR‐15b promote tumor progression and metastasis by suppressing key tumor‐suppressor proteins like RECK and PAQR3, and in lung adenocarcinoma its upregulation is linked to cisplatin resistance via repression of PEBP4."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nOutside the realm of oncogenesis, miR‐15b plays important roles in regulating differentiation, stress responses, and inflammation. In bone biology, miR‐15b promotes osteoblast differentiation by indirectly preserving Runx2 protein levels against Smurf1‐mediated degradation. In skin and aging models, reduced miR‐15b levels permit the upregulation of SIRT4, leading to mitochondrial dysfunction and senescence–associated secretory changes. Within the nervous system, miR‐15b attenuates neuroinflammatory responses during Japanese encephalitis virus infection by targeting RNF125 and modulating RIG‑I signaling, and it is implicated in mitigating α‑synuclein aggregation and toxicity in Parkinson’s disease via regulation of SIAH1. In diabetic conditions, altered miR‐15b expression influences pro‐fibrotic TGFβ signalling in the heart and modulates insulin signalling in retinal endothelial cells, while it also contributes to the control of angiogenesis via regulation of VEGFR‑2 expression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "14", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, miR‐15b modulates therapeutic responses and drug sensitivity across multiple tumor types. In models of chemotherapy‐resistant tongue squamous cell carcinoma and osteosarcoma, restoration of miR‐15b reverses the epithelial–mesenchymal transition and overcomes multidrug resistance—an effect linked to its targeting of factors such as BMI1 and Wee1. In gastric cancer, treatment with agents like dihydroartemisinin elevates miR‑15b expression, resulting in Bcl‑2 downregulation and enhanced apoptosis. Additionally, in colorectal cancer, the SIRT1/miR‑15b/ACOX1 axis has been shown to suppress metastatic behavior, while in the context of neuroblastoma, miR‑15b (in concert with related family members) downregulates MYCN to inhibit tumor growth."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "6"}, {"type": "fg_f", "ref": "21"}, {"type": "fg_f", "ref": "9"}, {"type": "fg_f", "ref": "22"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "L Sun, Y Yao, B Liu, et al.
