| HGNC Family | Non-coding RNAs |
| Name | microRNA 16-1 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Multiple studies have demonstrated that its downregulation in hematologic malignancies – such as chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL) and myelodysplastic syndromes – is strongly linked to the overexpression of anti‐apoptotic proteins like BCL2 and to altered cell cycle regulation (via cyclins and other regulators). In these settings, loss or deletion of miR‑16‑1 leads to enhanced proliferation of malignant B cells, defective apoptotic responses, and genomic instability. Furthermore, miR‑16‑1 is implicated in modulating cell‐cycle checkpoints, as it can influence the expression of key cell cycle drivers and is itself subject to regulation by transcription factors (e.g., E2F) and epigenetic modifiers (e.g., HDACs)."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in hematologic cancers, miR‑16‑1 governs multiple oncogenic signaling pathways in both hematologic and solid tumors. It directly targets several survival and proliferative genes – including MCL1, cyclin D1, cyclin E, and MYB – and is involved in the regulation of angiogenic factors such as VEGF and VEGF‑A. Altered miR‑16‑1 expression, often caused by epigenetic mechanisms (for example, by the actions of c‑Myc, HDAC3, or by competitive sponging via oncogenic long noncoding RNAs), contributes not only to unchecked cell proliferation and resistance to therapy but also to enhanced migratory and invasive properties. These defects promote aggressive disease features and drug resistance in cancers such as CLL, non‑small cell lung cancer, prostate cancer and osteosarcoma."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "12", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMoreover, emerging evidence in solid tumors and non‐cancer disorders highlights that miR‑16‑1 is not only a key regulator of tumor cell proliferation and motility but also modulates responses to external signals transmitted via exosomal communication. In prostate, breast, colorectal and lung cancers, as well as in conditions such as diabetic complications and placental dysfunction, reduced miR‑16‑1 expression is associated with enhanced epithelial‐mesenchymal transition, chemoresistance and aberrant activation of signaling pathways (including TGF‑β, Hedgehog and MAPK cascades).
