| HGNC Family | Non-coding RNAs |
| Name | microRNA 16-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In multiple studies, altered expression of miR‑16 (including that originating from MIR16‑2) has been implicated in the modulation of cell cycle regulators, apoptosis, and differentiation. In contexts such as esophageal cancer and progenitor cell biology, aberrant miR‑16 expression correlates with deregulated tumor‐promoting pathways and impaired regenerative responses, underscoring a critical role in cellular homeostasis and disease pathogenesis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn malignant tumors, reduced expression or loss of miR‑16 derived from the MIR16‑2 cluster is frequently associated with enhanced proliferation, invasion, and chemoresistance. Similar tumor‑suppressive properties have been observed in colorectal cancer, gastric cancer, chordoma, and neuroblastoma, where exogenous restoration of miR‑16 (commonly measured as miR‑16‑5p) led to the inhibition of oncogenes such as ITGA2, SMAD3, and MYCN, ultimately reducing tumor cell proliferation and increasing sensitivity to chemotherapy."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "9"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond direct tumor suppression, miR‑16 (including products from MIR16‑2) has gained attention for its diagnostic and therapeutic potential. Its stable expression in exosomal preparations suggests utility as a circulating biomarker in cancers such as breast and glioma, while its modulation influences cancer cell radiosensitivity and apoptotic responses in cervical, lung, and prostate cancer models by targeting genes like CARM1, MEK1, and AKT3. In papillary thyroid and osteosarcoma cells, for example, oncogenic long non‑coding RNAs and circular RNAs act as molecular sponges to sequester miR‑16‑5p, thereby de‐repressing targets that promote tumor progression. Moreover, in non‐cancer settings—ranging from cardiomyocyte endoplasmic reticulum and oxidative stress to advancements in biomarker research for community‑acquired pneumonia—miR‑16’s functional modulation is linked to altered cellular stress responses and tissue homeostasis.
