MIR17 Gene

HGNC Family Non-coding RNAs
Name microRNA 17
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR17, a key microRNA embedded in the polycistronic miR‐17–92 cluster, plays multifaceted roles in cancer biology. Aberrant over‐expression of this cluster has been documented in various malignancies—including B‐cell lymphomas, retinoblastoma, pancreatic cancer, clear cell renal carcinoma, and hepatocellular carcinoma—where it cooperates with oncogenic drivers such as c‐Myc and E2F. In these settings, MIR17 contributes to enhanced proliferation and survival by directly targeting tumor suppressors (for example, PTEN and pro‐apoptotic factors like Bim) and cell cycle inhibitors (such as CDKN1A/p21), while engaging in complex positive and negative feedback loops with regulators like cyclin D1. Intriguingly, in specific breast cancer contexts MIR17 may act in a tumor‐suppressive manner by down‐regulating AIB1 and thereby attenuating estrogen receptor signaling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "12"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its oncogenic functions, MIR17 also modulates diverse aspects of cellular behavior including differentiation, migration, invasion, and stress responses. By targeting key regulators such as HBP1 and SMAD7, MIR17 can affect pathways including Wnt/β‐catenin signaling and fibronectin expression, thereby influencing epithelial–mesenchymal transitions and metastatic potential. Its expression is likewise dynamically regulated during aging and immune cell activation—for instance, altered MIR17 levels have been linked to T‐cell activation changes in multiple sclerosis and to shifts in mesenchymal stem cell lineage commitment that impact osteogenic differentiation. Moreover, in certain solid tumors and chemoresistant cellular models, MIR17 deregulation is associated with modifications in autophagy and endothelial adhesion, further underscoring its pivotal role in both malignant progression and tissue homeostasis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "13", "end_ref": "30"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMIR17’s influence is not confined solely to cancer. It is increasingly recognized as a modulator in non‐neoplastic contexts and as a candidate biomarker. Studies have implicated MIR17 in neuronal cell cycle arrest during differentiation and in the regulation of stem cell populations. Altered MIR17 expression contributes to nonmalignant disorders such as autosomal dominant polycystic kidney disease and non‐traumatic osteonecrosis, while long noncoding RNAs—including the MIR17 host gene (MIR17HG) and HOTAIR—further refine MIR17’s activity through competitive endogenous RNA interactions. Moreover, circulating MIR17 levels have garnered attention as potential diagnostic markers, and viral mechanisms have been described that selectively down‐regulate MIR17 to modulate host cell responses. Collectively, these observations underscore the promise of therapeutically targeting MIR17 or its regulatory network in a broad spectrum of disease settings."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "31", "end_ref": "35"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Lin He, J Michael Thomson, Michael T Hemann, et al.
