MIR185 Gene

HGNC Family Non-coding RNAs
Name microRNA 185
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMiR‐185 is emerging as a critical regulator of cancer cell behavior by functioning predominantly as a tumor suppressor. Numerous studies have demonstrated that miR‐185 attenuates proliferation, migration, invasion, and therapeutic resistance in various malignancies—including ovarian, prostate, breast, colorectal, hepatocellular, nasopharyngeal, non‐small cell lung, and glioma—by targeting a wide array of oncogenic pathways. For example, miR‐185 suppresses key drivers of cell cycle progression and metastasis by directly repressing factors such as Six1, RhoA, Cdc42, c‐Myc, and DNMT1; it also targets the androgen receptor and VEGF family members, thereby modulating both tumor growth and chemosensitivity. In several contexts, its downregulation correlates with tumor recurrence, metastasis, and radio‐/chemo‐resistance, while restoration of miR‐185 levels enhances cellular apoptosis and sensitizes tumors to anticancer treatments."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "23"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its direct antitumor effects, miR‐185 plays an important role in lipid and cholesterol homeostasis. In hepatic cells, miR‐185 downregulates the expression of regulators involved in cholesterol biosynthesis and uptake—such as SREBP‐2 and the scavenger receptor SR‐BI—as well as modulating LDL receptor levels via accessory factors. These actions not only affect lipid metabolism but also have implications for cardiovascular disease and ischemic stroke, with altered circulating miR‐185 levels serving as potential biomarkers for vascular dysfunction."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "24", "end_ref": "28"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nAdditionally, miR‐185 is implicated in diverse non‐neoplastic contexts. In the immune system, its reduced expression in 22q11.2 deletion syndrome contributes to aberrant T cell development through the dysregulation of targets such as Mzb1, Nfatc3, and Camk4. It also partakes in developmental and fibrotic processes—for instance, by targeting Dlx2 and modulating amelogenesis and osteogenesis in cleidocranial dysplasia, as well as influencing renal fibrosis via a DNMT1/MEG3 axis.
