MIR191 Gene

HGNC Family Non-coding RNAs
Name microRNA 191
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐191 is emerging as a multifunctional regulator with context‐dependent roles in tumorigenesis. In several cancers its aberrant overexpression promotes oncogenic properties through the direct targeting and down‐regulation of key tumor suppressors. For example, in hepatocellular carcinoma miR‐191 overexpression is associated with enhanced cell proliferation and decreased apoptosis, whereas in ovarian and prostate cancers the creation of a miR‐191 target site in the 3′‐UTR of MDM4 leads to translational inhibition that delays tumor progression. In breast cancer, estrogen‐dependent upregulation of the miR‐191/425 cluster has been shown to protect cancer cells from hormone deprivation–induced apoptosis and to modulate global gene expression by targeting regulators such as SATB1, DAB2 and DICER1; similarly, in colorectal and gastric carcinomas miR‐191 promotes invasiveness by directly repressing TIMP3 expression. In addition, miR‐191 can be induced by environmental factors – such as hypoxia in breast cells and arsenite exposure in lung and liver epithelial cells – thereby activating TGFβ, Wnt/β‐catenin and HIF‐dependent pathways that enhance migration, epithelial–mesenchymal transition and acquisition of stem‐like properties. Furthermore, in endometriosis and its progression to ovarian malignancy, miR‐191 targets DAPK1, contributing to the malignant transformation process, while overexpression in cholangiocarcinoma further underscores its role in suppressing key epigenetic regulators (e.g. TET1) that modulate p53."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "16"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its mechanistic roles in promoting malignancy, miR‐191 holds promise as a diagnostic and prognostic biomarker across diverse clinical settings. Its elevated levels in body fluids have been correlated with disease severity in traumatic brain injury, colon adenocarcinoma and bladder cancer, and its dysregulation is further implicated in modulating cellular senescence in keratinocytes.
