MIR196B Gene

HGNC Family Non-coding RNAs
Name microRNA 196b
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nExtensive studies have revealed that miR‑196b plays a multifaceted role in solid tumors where its expression is frequently deregulated and functionally linked to cancer progression. In colorectal, lung, gastric, pancreatic, cervical, and head and neck cancers, miR‑196b modulates key cellular processes including stemness, proliferation, migration, invasion, chemoresistance, and metastases. For instance, in colorectal cancer, up‐regulation of this miRNA has been shown to enhance stem cell properties and chemoresistance through targeting negative regulators of STAT3 signaling, while in non‑small cell lung cancer it promotes cell migration and proliferation by directly repressing tumor suppressors such as FAS, GATA6, and TSPAN12. Moreover, in other tumors such as gastric cancer and cervical cancer, aberrant miR‑196b levels influence oncogenic pathways involving homeobox genes and angiogenic regulators, underscoring a context‑dependent function for miR‑196b in tumor biology."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "17"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn hematological malignancies, miR‑196b exhibits complex and context‑dependent functions. In MLL‑rearranged leukemias and other acute leukemias, miR‑196b is often highly expressed and its ectopic expression modulates oncogene–tumor suppressor networks by directly targeting genes such as HOXA9, MEIS1, and the proapoptotic factor FAS. Experimental evidence from primary bone marrow transplantation studies and patient samples indicates that miR‑196b can delay or accelerate leukemogenesis depending on which targets (oncogenic or tumor‐suppressive) are predominantly affected. These findings highlight an intricate regulatory balance whereby miR‑196b orchestrates key transcriptional programs during normal hematopoiesis as well as in leukemic transformation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "18", "end_ref": "29"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nEmerging evidence also underscores novel regulatory layers and clinical applications associated with miR‑196b. In addition to being subject to epigenetic modifications—such as promoter hypomethylation that can account for its aberrant overexpression—the miRNA is involved in regulatory interactions with long noncoding RNAs and can be modulated by viral as well as inflammatory signals. For example, in the context of viral infections (including HIV and Ebola) and cigarette‑smoke–related pathologies, miR‑196b has been implicated in modulating host responses, while its circulating levels have shown promise as non‑invasive biomarkers for early cancer detection and differential diagnosis in conditions like thyroid nodules and laryngeal carcinomas. Moreover, recent work has indicated that factors such as ETS2 can repress miR‑196b transcription in certain gastric cancers, and novel circRNA networks may function as sponges for miR‑196b in acute myeloid leukemia, collectively offering new avenues for targeted therapy and improved prognostic assessment."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "30", "end_ref": "38"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Yu-Lun Liao, Ling-Yueh Hu, Kuo-Wang Tsai, et al.
