MIR199A2 Gene

HGNC Family Non-coding RNAs
Name microRNA 199a-2
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR199A2 is a critical genomic locus that encodes miR‐199a, which is processed into two mature species (miR‐199a‐3p and miR‐199a‐5p) that play multifaceted roles in cell biology. In ovarian cancer, for example, miR‐199a regulates inflammatory and protumor pathways by directly targeting IKKβ and by modulating NF‐κB and PTEN/AKT signaling; it also contributes to cancer stem cell differentiation and tissue homeostasis, as evidenced by its roles in processes such as myometrial quiescence. These findings underscore an important function for MIR199A2 in the regulation of “stemness” and differentiation in both malignant and normal tissues."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "5"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn diverse malignancies, MIR199A2 exhibits context‐dependent functions that can either restrain or promote tumor progression. In hepatocellular carcinoma, miR‐199a‐3p reduces cell proliferation and chemoresistance by downregulating targets such as CD44 and RGS17, while re‐expression in osteosarcoma suppresses cell growth and migration. In gastric and colorectal cancers, aberrant expression of miR‐199a correlates with enhanced metastasis, and in cutaneous squamous cell and breast cancers, miR‐199a controls key regulators of the epithelial–mesenchymal transition (including E‐cadherin and PAK4). These studies collectively reveal that miR‐199a plays a pivotal role in modulating oncogenic signaling, invasion, and drug sensitivity."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "6", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMIR199A2 also influences the tumor microenvironment and cellular metabolism. In endothelial cells, miR‐199a‐5p restricts angiogenic responses by directly targeting transcription factors such as Ets‐1, while in multiple myeloma, its re‐expression decreases GRP78 levels and overcomes chemoresistance. Furthermore, miR‐199a is involved in endoplasmic reticulum stress responses in hepatic cells and contributes to the metabolic reprogramming of hepatocellular carcinoma through hypoxia‐responsive mechanisms. A role in stromal modulation is evident from studies showing that exosome‐mediated transfer of miR‐199a in pancreatic stellate cells and in ischemic stroke models regulates fibrogenesis and cell–cell interactions, thereby emphasizing its importance in non–cell autonomous regulation within the tumor milieu."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "20", "end_ref": "28"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its established roles in cancer, altered MIR199A2 expression is emerging as a promising biomarker and functional modulator in several non‐malignant conditions. Elevated miR‐199a levels have been linked to chondrocyte senescence in osteoarthritis, while aberrant circulating levels serve as diagnostic indicators in early gastric and colorectal cancers. In testicular germ cell tumors, miR‐199a‐3p targets the de novo methyltransferase DNMT3A to restore tumor suppressor gene expression, and its impact on monocyte/macrophage differentiation suggests roles in immune regulation.
