MIR200A Gene

HGNC Family Non-coding RNAs
Name microRNA 200a
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR200A is emerging as a critical regulator of epithelial‐to‐mesenchymal transition (EMT) and metastasis in a wide spectrum of cancers. By directly targeting key EMT‐inducing transcriptional repressors such as ZEB1, ZEB2, and SIP1—and by dampening signaling molecules like β‐catenin—MIR200A helps to restore E‐cadherin expression, thereby maintaining an epithelial phenotype, reducing cell motility, and limiting invasive behavior. In several tumor types, MIR200A also governs the stem‐like cellular state and modulates immune evasion (for example, through the regulation of PD‐L1), thus exerting potent tumor‐suppressive effects."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its role in blocking EMT, MIR200A modulates several key cell signaling and cell cycle pathways that impact tumor progression. It has been shown to target p38α—thereby influencing redox regulation and the cellular oxidative stress response—and to repress Keap1 expression, which in turn activates the antioxidant Nrf2 pathway. Moreover, by regulating SIRT1 levels, MIR200A contributes to cell cycle arrest (via upregulation of p27 and downregulation of cyclin‐dependent kinases) and enhances the sensitivity of cancer cells to chemotherapeutic agents, as observed in ovarian and breast carcinomas."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "11"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nMIR200A also participates in complex regulatory networks that involve long non‐coding RNAs and microenvironmental factors. For example, in hepatocellular carcinoma, the oncogenic lncRNA HULC sequesters the MIR200A‐3p pathway to facilitate ZEB1‐induced EMT and metastasis, while in colorectal cancer the lncRNA H19 competes with MIR200A to derepress β‐catenin, thereby promoting tumor cell proliferation. In other settings, low MIR200A levels are linked to the activation of cancer‐associated fibroblasts, aberrant Notch signaling, and the formation of vasculogenic-like networks in triple‐negative breast cancer. Furthermore, altered MIR200A expression has been associated with prognostic outcomes in cancers of the nasopharynx, ovary, prostate, and even in endometrial and intestinal fibrotic disorders."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}, {"type": "fg_fs", "start_ref": "12", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Limo Chen, Don L Gibbons, Sangeeta Goswami, et al.
