| HGNC Family | Non-coding RNAs |
| Name | microRNA 206 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. Disruption of the encoded miRNA has been implicated in multiple skeletal muscle disorders, including amyotrophic lateral sclerosis (ALS) and Duchenne muscular dystrophy (DMD), as well as in several cancers. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Aug 2017] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‐206 has emerged as a multifunctional regulator with prominent tumor‐suppressive roles in diverse cancers. In breast and endometrial carcinomas, miR‐206 downregulates estrogen receptor‑α expression and its coregulators, while in gastric cancer it targets cyclin D2 to arrest cell cycle progression. In sarcomas such as rhabdomyosarcoma, it suppresses oncogenic drivers like c‑Met to inhibit cell proliferation and migration. Similarly, in lung cancers, miR‐206 downregulates c‑Met and Bcl2, thereby blocking epithelial–mesenchymal transition (EMT) and reducing invasiveness. Its activity has also been linked to breast cancer metabolic reprogramming via regulation of glycolytic enzymes and, in osteosarcoma, low miR‑206 expression correlates with poor prognosis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "14"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to these antitumor effects, miR‐206 plays critical roles in metabolic reprogramming and differentiation. Under conditions of sustained NRF2 signaling, miR‑206 levels are attenuated, which facilitates the redirection of glucose flux toward the pentose phosphate pathway; conversely, restoring miR‑206 expression rebalances metabolic gene expression. In the central nervous system, miR‑206 regulates brain‐derived neurotrophic factor (BDNF), thereby influencing memory function in Alzheimer’s disease. Its high, muscle‐specific expression is essential during myogenic differentiation, and in neuromuscular disorders such as amyotrophic lateral sclerosis, elevated circulating miR‑206 may serve as a compensatory response and a potential biomarker."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "15", "end_ref": "19"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its intrinsic effects on tumor cell behavior, miR‑206 modulates the tumor microenvironment and chemotherapeutic responses. By targeting Notch3, miR‑206 contributes to pro‐apoptotic signaling and modulates histone acetylation in certain hematologic malignancies. In lung cancer cells, overexpression of miR‑206 attenuates HGF‑induced EMT and angiogenesis via suppression of the c‑Met/PI3K/AKT/mTOR cascade.
