MIR215 Gene

HGNC Family Non-coding RNAs
Name microRNA 215
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]

Several studies demonstrate that following genotoxic stress, p53‐dependent induction of miR‐215 leads to cell cycle arrest by coordinately down‐regulating multiple transcripts—including thymidylate synthase, dihydrofolate reductase, DTL, and other checkpoint regulators—thereby exerting tumor‐suppressive effects and promoting a more differentiated, less “stem‐like” state in cancers such as colorectal carcinoma and renal cell carcinoma."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its cell‐cycle regulatory functions, miR‐215 also modulates chemotherapeutic responses by directly targeting key enzymes of nucleotide metabolism. By suppressing thymidylate synthase, dihydrofolate reductase, and related mediators, miR‐215 can influence sensitivity to antimetabolite chemotherapy. Depending on the cellular context, its altered expression may contribute to chemoresistance or, conversely, enhance apoptosis and cell cycle arrest in cancers including colorectal carcinoma, gastric cancer, and multiple myeloma."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "13"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond cell proliferation and chemotherapy response, miR‐215 is a pivotal regulator of tumor invasion and metastasis. It can affect cell migration and epithelial–mesenchymal transition by targeting key molecules such as the retinoblastoma tumor suppressor and transcriptional regulators including SRY‐Box factors. In hepatocellular carcinoma, for example, miR‐215 targets oncogenic lncRNAs (e.g. PCAT‐1 and FTX) to control downstream effectors like CRKL, whereas in other contexts it modulates cell polarity and tight junction formation via targets such as RAB11‐FIP2. In addition, aberrant miR‐215 expression has now been implicated not only in multiple tumor types (including glioma, ovarian cancer, and cervical cancer) but also in non–malignant conditions such as diabetic nephropathy, Hirschsprung disease, acute myeloid leukemia, ischemic stroke, and even viral infections.
