| Name | microRNA 2276 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nAlthough the query referenced MIR2276, the provided studies focus on the role of NKD2 in modulating Wnt signaling pathways. In one study investigating osteosarcoma, NKD2 expression was found to be significantly downregulated in metastatic and recurrent tumors. This decrease in NKD2, a known negative regulator of Wnt signaling, was associated with enhanced cell proliferation, migration, invasion, and overall tumor aggressiveness. Importantly, overexpression of NKD2 in metastatic osteosarcoma cell models not only curtailed these malignant properties in vitro but also substantially reduced tumor growth and metastasis in vivo, suggesting a potential therapeutic target for intervention."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nA complementary study explored the regulation of NKD2 from a developmental perspective in a mouse model. Here, NKD2, an EF hand protein acting as an inducible antagonist of the Wnt (Wg) signaling, was demonstrated to be a direct transcriptional target of the homeobox gene Hoxc8. Techniques including chromatin immunoprecipitation, electrophoretic mobility shift assays, and reporter assays confirmed that endogenous Hoxc8 binds the enhancer of NKD2, leading to suppression of its expression. This finding introduces a novel mechanism for cross-talk between Hoxc8-driven differentiation pathways and Wnt-mediated cellular processes."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nTogether, these studies underscore the critical function of NKD2 as a negative regulator of Wnt signaling, with significant implications in both tumor suppression in osteosarcoma and developmental regulation via Hoxc8. Although MIR2276 was mentioned in the query, no direct evidence regarding its role was provided in these abstracts. Instead, the insights on NKD2 highlight its dual impact on tumor progression and cellular differentiation, thereby illuminating a key molecular axis that could be exploited for targeted therapeutic strategies."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "S Zhao, L Kurenbekova, Y Gao, et al.
