| HGNC Family | Non-coding RNAs |
| Name | microRNA 24-2 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

Early studies established that small regulatory RNAs govern many cellular processes, with MIR24-2 itself being subject to complex transcriptional and epigenetic regulation—including promoter DNA methylation and SP1–mediated transcription—that sets its baseline expression in normal cells and influences its incorporation into multi‐miRNA clusters. These mechanisms provide a platform for context‐specific regulation of downstream targets that shape responses to cellular stress and control cell fate decisions."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "7"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn cancer and other disease models, the mature product(s) of MIR24-2 (commonly detected as miR-24-3p) display context-dependent roles that can be either oncogenic or tumor suppressive. In several malignancies—including glioma, breast, bladder, nasopharyngeal, lung, hepatocellular, and pancreatic cancers—as well as in hematologic and immune disorders, miR-24-3p has been shown to modulate key cellular functions. It regulates proliferation, apoptosis, autophagy, invasion, epithelial-to-mesenchymal transition, and immune cell crosstalk by targeting a myriad of mRNAs such as those encoding H2AX, FGF11, ATG4A, DEDD, ST7L, p27^Kip1, p53, and Bim. Moreover, MIR24-2 influences additional physiological processes (for example, aspects of smooth muscle and phagocyte function, as well as natural killer cell activity) by altering the expression of cell cycle regulators and apoptosis mediators. Such pleiotropic actions underscore its role in modulating cell survival pathways, metastatic behavior, and responses to external stress—including the effects of hypoxia and cytotoxic drugs."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "8", "end_ref": "31"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBuilding on these mechanistic insights, emerging strategies are exploring the modulation of MIR24-2 expression—either by inhibiting its aberrant elevation or by its replacement in contexts where it is diminished—to overcome chemoresistance and to reprogram dysregulated pathways in cancer. Notably, exosomal transfer of miR-24-3p from stromal or cancer-associated cells further highlights its role in intercellular communication and the shaping of tumor microenvironments.
