MIR26A1 Gene

HGNC Family Non-coding RNAs
Name microRNA 26a-1
Description microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009]
Summary
{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMIR26A1, which produces miR‑26a, acts as a critical regulator in cancer. In many tumor types—including hepatocellular, nasopharyngeal, breast, gastric, bladder, renal, lymphoma, glioma, and others—miR‑26a is frequently downregulated, leading to unchecked expression of oncogenic targets such as EZH2, MTDH, MCL‑1, FGF9, and components of the Notch pathway. In these settings, restoration of miR‑26a expression can induce cell‑cycle arrest, promote apoptosis, and reduce invasion and metastasis. In contrast, in certain contexts such as lung cancer, elevated levels of miR‑26a have been associated with enhanced migratory and invasive capabilities via PTEN suppression and AKT activation. Moreover, oncogenic drivers (e.g., MYC) and disruptions in miRNA maturation contribute to its dysregulation, thereby modulating chemoresistance and tumor progression."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "20"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its roles in oncogenesis, MIR26A1 influences several non‑neoplastic processes. miR‑26a modulates key inflammatory and metabolic pathways by regulating cytokine production (including interleukin‑6 and interferon‑β), controlling TGF‑β/SMAD signaling, and fine‑tuning NF‑κB activity. These actions affect endothelial function, vascular smooth muscle cell proliferation and differentiation, angiogenesis, and wound healing. In metabolic tissues, altered miR‑26a levels have been linked to improved insulin sensitivity, reduced hepatic glucose production, and attenuated fibrogenesis, while in conditions such as pulmonary fibrosis and diabetic wound repair, manipulation of miR‑26a expression can markedly shift the balance between repair and pathologic remodeling."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "21", "end_ref": "35"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nGenetic studies further link variations in the pri‑MIR26A1 transcript to heightened risk for premalignant lesions and altered therapeutic outcomes, suggesting that miR‑26a holds promise as a diagnostic and prognostic biomarker. Additionally, miR‑26a is involved in regulating neurotrophic factors and neuronal signaling, while its own expression is subject to regulation by other miRNAs and RNA‑binding proteins. These findings highlight a broader regulatory network in which MIR26A1 participates, with implications that extend from tumorigenesis and inflammation to neurobiology."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "36", "end_ref": "39"}]}, {"type": "t", "text": "\n"}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Sandrine Sander, Lars Bullinger, Kay Klapproth, et al.
