| HGNC Family | Non-coding RNAs |
| Name | microRNA 29b-1 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In the context of neurodegenerative disease, loss or down‐regulation of the miR‐29a/b-1 cluster leads to derepression of key targets such as BACE1, thereby promoting excessive amyloid‐β generation in Alzheimer’s disease. Moreover, an unusual hexanucleotide motif in miR‑29b directs its nuclear localization, suggesting additional layers of posttranscriptional and epigenetic regulation not shared by typical cytoplasmic miRNAs."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn numerous cancers, miR‑29b functions predominantly as a tumor suppressor by directly targeting a spectrum of oncogenic and epigenetic regulators. It down‐regulates DNA methyltransferases (DNMT1, DNMT3A, and DNMT3B) and their transactivators (e.g. Sp1), thereby restoring the expression of tumor suppressor genes and reversing aberrant DNA hypermethylation seen in acute myeloid leukemia, chronic lymphocytic leukemia, and gastric cancer. In solid tumors such as hepatocellular, non–small-cell lung, prostate, colorectal, pancreatic, and certain head and neck carcinomas, miR‑29b suppresses cell proliferation, migration, invasion, and angiogenesis by targeting key molecules such as ABL1, MMP2, LAMC1, Akt3, and by interfering with signaling cascades (e.g. PI3K/AKT, MAPK). Its loss correlates with enhanced extracellular matrix deposition, epithelial-to-mesenchymal transition and chemoresistance, while enforced expression restores apoptotic responses and diminishes invasive behavior. Complementary studies further reveal that miR‑29b is embedded in intricate regulatory networks involving long noncoding RNAs (such as HOTAIR and H19) and transcription factors (including c-Myc and KLF4), which in turn fine-tune its expression and downstream effects in malignancies."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "3", "end_ref": "25"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncogenesis, miR‑29b plays a pivotal role in regulating extracellular matrix remodeling and fibrotic processes in non-malignant conditions. In cardiovascular and endometrial fibrosis, as well as in liver fibrogenesis and osteoarthritis, miR‑29b modulates the expression of key structural proteins such as collagens and matrix metalloproteinases, thereby attenuating pathologic fibrotic deposition and preserving tissue homeostasis. In addition, by targeting regulators like progranulin in chondrocytes, it promotes apoptosis and counteracts joint degeneration.
