| HGNC Family | Non-coding RNAs |
| Name | microRNA 300 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nMicroRNA‐300 (miR‑300) has emerged as a critical regulator of epithelial–mesenchymal transition (EMT), a pivotal process in tumor metastasis. In certain cancers, such as those described in studies on tumor cell metastasis and hepatocellular carcinoma, miR‑300 modulates EMT by influencing key signaling pathways—including FAK/PI3K/AKT—and can reverse TGF‑β1–induced EMT. Notably, in colorectal cancer contexts, miR‑300 even appears to promote EMT via a p53‐dependent mechanism, underscoring a context‐dependent role in EMT regulation."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "3"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn several malignancies miR‑300 acts as a tumor suppressor. For instance, studies in pituitary tumors and glioblastoma demonstrate that miR‑300—by targeting oncogenic mediators such as PTTG1 and ROCK1—suppresses cell proliferation, migration, and invasion. Similarly, investigations in bladder cancer and non‑small cell lung cancer show that restoration of miR‑300 expression impedes tumor cell motility by downregulating targets like SP1 and HIF3α, thereby contributing to reduced metastatic potential."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "4", "end_ref": "7"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nConversely, miR‑300 has also been reported to exert oncogenic functions in a subset of cancers. In colorectal and liver cancers, elevated miR‑300 levels have been linked to enhanced proliferation and metastasis. Mechanistically, in these settings miR‑300 targets tumor suppressors such as p53 and FOXO1 while activating downstream pro‑survival and cell cycle pathways (including AKT/mTOR), thereby fostering an environment conducive to tumor progression. In addition, in lung cancer cells exposed to ionizing radiation, miR‑300 desensitizes the DNA damage response by suppressing p53 and apaf1, further underlining its role in promoting radioresistance."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "3"}, {"type": "fg_f", "ref": "8"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond its dichotomous roles in tumor cells, miR‑300 influences additional aspects of cancer biology and non‑malignant conditions. In glioma stem‑like cells, for example, miR‑300 is upregulated to enhance self‑renewal through direct targeting of LZTS2, while in gastric cancer it suppresses oncogenic drivers such as UBE2C. In chronic myelogenous leukemia, miR‑300 modulates cell quiescence and impairs natural killer cell antitumor activity via PP2A activation. Its impact extends to inflammatory and degenerative conditions as well: elevated circulating miR‑300 levels have been observed in acute lung injury—where its targeting of IκBα promotes inflammatory factor expression—and alterations in its serum levels are associated with diseases like knee osteoarthritis and multiple sclerosis. Finally, studies in osteosarcoma, prostate cancer, melanoma, and papillary thyroid cancer reveal that miR‑300 regulates invasion, metastasis, and cell cycle progression by modulating targets such as Twist1, DAB1, and components of the TP53/GADD45B axis."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "10", "end_ref": "22"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these studies underscore the complex, context‑dependent functions of miR‑300 in human diseases. By targeting diverse molecular effectors—including PTTG1, ROCK1, SP1, p53, FOXO1, LZTS2, UBE2C, and others—miR‑300 can either suppress or promote tumorigenesis and metastasis, regulate cell cycle progression and apoptosis, modulate DNA damage responses, influence stemness and quiescence, and even alter inflammatory signaling. These multifaceted roles render miR‑300 not only a promising biomarker for disease diagnosis and prognosis but also a potential therapeutic target whose modulation may improve treatment outcomes across a range of malignancies and other pathological conditions."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "23"}, {"type": "fg_f", "ref": "2"}]}, {"type": "t", "text": "\n"}]}]}]}]}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Jingshuang Yu, Furong Xie, Xin Bao, et al.
