| HGNC Family | Non-coding RNAs |
| Name | microRNA 30d |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

{"type": "root", "children": [{"type": "p", "children": [{"type": "t", "text": "\nmiR‑30d is a multifunctional microRNA that plays diverse and sometimes opposing roles in cancer. In several malignancies – including melanoma, prostate, breast, non‑small cell lung, thyroid and other human cancers – miR‑30d has been found to either promote progression or act as a tumor suppressor. For instance, studies have shown that in melanoma its upregulation facilitates metastasis and immunosuppression by directly targeting enzymes involved in glycosylation and altering cytokine production, while in other tumors it negatively regulates the tumor suppressor p53 and modulates the expression of oncogenes such as SOCS1, cyclin E2, metadherin and GRP78. Moreover, miR‑30d impacts cell cycle progression, apoptosis, autophagy and chemoresistance through direct targeting of key factors (e.g. beclin‑1) in these processes. These findings underscore the complex role of miR‑30d in regulating tumor cell proliferation, invasion and survival."}, {"type": "fg", "children": [{"type": "fg_fs", "start_ref": "1", "end_ref": "25"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nBeyond oncogenesis, miR‑30d is implicated in a variety of non‑malignant processes. In reproductive biology, for example, endometrial epithelial cells upregulate and release miR‑30d—packaged in exosomes—to modulate gene expression in pre‑implantation embryos and promote trophoblast adhesion, thereby contributing to endometrial receptivity. Altered circulating or urinary levels of miR‑30d have also been proposed as sensitive non‑invasive biomarkers for conditions such as acute myocardial infarction, kidney injury and even as markers predictive of cardiac resynchronization therapy response. In addition, dysregulation of miR‑30d has been associated with abnormal extracellular matrix remodeling in pelvic supportive tissues, as well as in trophoblast function in gestational diabetes, emphasizing its broader role in tissue homeostasis and repair."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "5"}, {"type": "fg_fs", "start_ref": "26", "end_ref": "28"}, {"type": "fg_f", "ref": "17"}, {"type": "fg_fs", "start_ref": "29", "end_ref": "35"}]}, {"type": "t", "text": "\n"}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nIn addition to its direct effects on cell behavior, miR‑30d is integrated within complex regulatory networks that fine‑tune cellular responses. Its transcription is modulated by several key signaling pathways—including TGF‑β/Smad, PI3K/Akt/FOXO and hypoxia‐responsive mechanisms—and it is subject to regulation by competing endogenous RNAs such as long non‑coding RNAs.
