| Name | microRNA 3134 |
| Description | microRNAs (miRNAs) are short (20-24 nt) non-coding RNAs that are involved in post-transcriptional regulation of gene expression in multicellular organisms by affecting both the stability and translation of mRNAs. miRNAs are transcribed by RNA polymerase II as part of capped and polyadenylated primary transcripts (pri-miRNAs) that can be either protein-coding or non-coding. The primary transcript is cleaved by the Drosha ribonuclease III enzyme to produce an approximately 70-nt stem-loop precursor miRNA (pre-miRNA), which is further cleaved by the cytoplasmic Dicer ribonuclease to generate the mature miRNA and antisense miRNA star (miRNA*) products. The mature miRNA is incorporated into a RNA-induced silencing complex (RISC), which recognizes target mRNAs through imperfect base pairing with the miRNA and most commonly results in translational inhibition or destabilization of the target mRNA. The RefSeq represents the predicted microRNA stem-loop. [provided by RefSeq, Sep 2009] |

In studies using breast cancer cell lines, overexpression of MIR3134 led to a marked 4–8‐fold increase in the levels of ARE‐bearing mRNAs—including key regulators such as SOX9, VEGFA, and EGFR—by stabilizing these transcripts rather than by affecting transcription, which is evident from its resistance to actinomycin D treatment."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nDetailed analyses employing quantitative RT-PCR and 3′UTR luciferase assays further confirmed that this upregulation is sequence specific, as mutations in MIR3134 diminish its stabilizing effect on target transcripts. Additionally, comparative overexpression studies with the ARE-binding protein HuR have revealed that there is extensive overlap between the sets of genes regulated by both MIR3134 and HuR, indicating a cooperative mechanism; indeed, wild-type MIR3134—but not its mutated counterpart—promotes stabilization of HuR-regulated transcripts, as validated by RNA-immunoprecipitation experiments."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "t", "text": "\n\n"}, {"type": "p", "children": [{"type": "t", "text": "\nCollectively, these findings position MIR3134 as a positive regulator in post-transcriptional control, where it collaborates with canonical ARE-binding proteins to enhance mRNA stability—a mechanism that stands in contrast to the more typical microRNA-mediated repression. This alternative mode of action may have broad implications for the regulation of cellular processes such as proliferation, angiogenesis, and possibly tumorigenesis."}, {"type": "fg", "children": [{"type": "fg_f", "ref": "1"}]}, {"type": "t", "text": ""}]}, {"type": "rg", "children": [{"type": "r", "ref": 1, "children": [{"type": "t", "text": "Shivani Sharma, Suneer Verma, Madavan Vasudevan, et al.
